The immune response of NK cells is mediated by both activation and inhibitory signals mediated by cell surface receptors. Phosphorylation of the immunoreceptor tyrosine inhibitory motif (ITIM) in the cytosolic region of the inhibitory receptor recruits tyrosine or lipid phosphatases to modulate intracellular signaling molecules that are activated by Syk, ZAP70 or PI 3 kinase pathways. Recent studies on a number of null mice, such as Syk and ZAP70, have shown that NK cells possess a broad and robust receptor system that guarantees their normal development and exerts their effects. This article intends to review the progress of NK cell signaling research.
