目的 探讨人巨细胞病毒包涵体的组成及意义。方法 采用显微切割技术结合PCR和Southern杂交及免疫组织化学的催化信号扩增法 ,检测人巨细胞病毒的核酸和 3种不同阶段表达的抗原。结果 通过液压显微切割系统将组织中巨细胞病毒包涵体游离出来 ,PCR扩增出预期长度的DNA片段 ,Southern杂交证实为HCMV的核酸片段 ,催化信号扩增法检测结果显示包涵体主要表达立即早期和早期抗原DDG9/CCH2 ,而基质蛋白AAC10则为阴性。结论 人巨细胞病毒包涵体的成分包含有病毒DNA和特异阶段表达的抗原产物
Objective To investigate the composition and significance of human cytomegalovirus inclusion bodies. Methods The nucleic acids of human cytomegalovirus (CMV) and the antigens expressed in three different stages were detected by microdissection combined with PCR and Southern blotting and immunohistochemistry. Results The inclusion body of cytomegalovirus was dissociated from the tissue by hydraulic microdissection system. The DNA fragment of expected length was amplified by PCR. The Southern hybridization confirmed that it was the nucleic acid fragment of HCMV. The result of catalytic signal amplification showed that the inclusion body was mainly expressed immediately The early and early antigens DDG9 / CCH2, while the matrix protein AAC10 was negative. Conclusion The composition of human cytomegalovirus inclusion body contains viral DNA and specific stage expressed antigen product
