探讨直接利用心肌匀浆测定肌浆网Ca2+-ATP酶的方法,并观察左室肥厚时肌浆网Ca2+-ATP酶的变化。采用p-硝苯基磷酸(p-NPP)法建立SD大鼠心肌匀浆肌浆网Ca2+-ATP酶检测的最佳条件;取6周龄自发性高血压大鼠(SHR)及对照组饲养24周,观察血压、左室肥厚及心肌肌浆网Ca2+-ATP酶活力。结果在心肌匀浆中建立了肌浆网Ca2+-ATP酶活力测定的最佳条件;并发现SHR左室肥厚时肌浆网Ca2+-ATP酶活力较对照组明显下降(P<0.01)。
To explore the direct use of myocardial homogenate sarcoplasmic reticulum Ca2 + -ATP enzyme method and observed sarcoplasmic reticulum Ca2 + -ATP enzyme changes in left ventricular hypertrophy. The optimal conditions of sarcoplasmic reticulum Ca2 + -ATPase in SD rat myocardial tissue were established by p-nitrophenyl phosphate (p-NPP) method. Six-week-old spontaneously hypertensive rats (SHR) and control group were bred After 24 weeks, blood pressure, left ventricular hypertrophy and cardiac sarcoplasmic reticulum Ca2 + -ATPase activity were observed. Results The optimum condition of sarcoplasmic reticulum Ca2 + -ATPase activity was established in myocardial homogenate. It was found that Ca2 + -ATPase activity in sarcoplasmic reticulum was significantly decreased in SHR group compared with control group (P <0.01).
