目的观察氧糖剥夺后处理在缺氧缺糖-再灌注损伤中的抗凋亡作用。方法应用SK-N-SH细胞进行氧糖剥夺4h,然后恢复氧糖20h建立氧糖剥夺-再灌注损伤模型,3次循环的氧糖剥夺10min/复氧糖10min进行氧糖剥夺后处理;细胞分为对照组、氧糖剥夺-再灌注组(OGD/R组)与后处理组,MTT法与Hoechst33258染色分别检测氧糖剥夺后处理对细胞存活率与凋亡的影响。结果 OGD/R组细胞存活率较对照组下降约43%(P<0.01),后处理组细胞存活率比OGD/R组提高约22%(P<0.01);凋亡计数显示,OGD/R组和后处理组的细胞凋亡率较对照组增高(P<0.01),而后处理细胞凋亡率低于OGD/R组(P<0.01)。结论氧糖剥夺后处理能提高氧糖剥夺-再灌注损伤细胞的存活率,可能与其抑制细胞凋亡有关。
Objective To observe the anti-apoptotic effect of post-oxygen glucose deprivation (OGD) treatment on oxygen-glucose deprivation-reperfusion injury. Methods Oxygen-glucose deprivation was performed for 4 h in SK-N-SH cells, then oxygen glucose deprivation-reperfusion injury model was established after oxygen glucose recovery for 20 h. Oxygen-glucose deprivation for 3 cycles was performed for 10 min and reoxygenation for 10 min. The rats were divided into control group, OGD / R group and postconditioning group. MTT assay and Hoechst33258 staining were used to detect the effects of post-OGD treatment on cell viability and apoptosis. Results The survival rate of OGD / R group was about 43% lower than that of control group (P <0.01). The survival rate of OGD / R group was about 22% higher than that of OGD / R group (P <0.01) The apoptotic rate of the treated group and the post-treated group was higher than that of the control group (P <0.01), while the apoptosis rate of the post-treatment group was lower than that of the OGD / R group (P <0.01). Conclusion Oxygen-glucose deprivation can improve the survival rate of oxygen-glucose deprivation-reperfusion injury cells, which may be related to its inhibition of apoptosis.
