目的探讨Th1和Th2细胞特异性转录因子T-bet/GATA-3在梅毒血清固定患者免疫失衡中的作用。方法采用实时荧光定量PCR检测30例梅毒血清固定患者,22例潜伏梅毒患者和30例正常对照者外周血单个核细胞中T-bet,GATA-3 mRNA的表达水平,采用流式细胞术检测Th1,Th2细胞比例。结果梅毒血清固定组、潜伏梅毒组T-bet mRNA表达水平及T-bet/GATA-3比值明显低于正常对照组,差异有统计学意义(P均<0.05)。而GATA-3 mRNA表达水平在各组之间差异均无统计学意义(P>0.05)。血清固定组、潜伏梅毒组与正常对照组比较,Th1细胞比例明显降低,Th2细胞比例明显升高,Th1/Th2比值明显下降,差异有统计学意义(P<0.05)。T-bet/GATA-3比值与Th1/Th2比值呈正相关(P<0.05);T-bet,GATA-3 mRNA表达水平,Th1,Th2细胞比例与血清RPR滴度之间不存在相关性(P均>0.05)。结论梅毒血清固定患者存在T-bet/GATA-3表达失衡,主要与转录因子T-bet的低表达有关,T-bet/GATA-3表达失衡可能在梅毒血清固定形成中起重要作用。
Objective To investigate the role of Th1 and Th2 cell-specific transcription factor T-bet / GATA-3 in immune imbalance in patients with syphilis sera. Methods The expression of T-bet and GATA-3 mRNA in peripheral blood mononuclear cells from 30 cases of syphilis, 22 cases of latent syphilis and 30 cases of normal controls were detected by real-time fluorescence quantitative PCR. The levels of Th1 , Th2 cell ratio. Results The T-bet mRNA expression and T-bet / GATA-3 ratio in syphilis serum group and latent syphilis group were significantly lower than those in normal control group (all P <0.05). However, there was no significant difference in GATA-3 mRNA expression among the groups (P> 0.05). Compared with normal control group, the proportion of Th1 cells in serum-fixed group and latent syphilis group was significantly decreased, the proportion of Th2 cells was significantly increased, and the Th1 / Th2 ratio was significantly decreased (P <0.05). The ratio of T-bet / GATA-3 was positively correlated with Th1 / Th2 ratio (P <0.05). There was no correlation between the expression of T-bet, GATA-3 mRNA and the ratio of Th1 and Th2 cells All> 0.05). Conclusion The imbalance of T-bet / GATA-3 expression in syphilis serum is mainly associated with the low expression of transcription factor T-bet. The imbalance of T-bet / GATA-3 expression may play an important role in the formation of syphilis serum.
