目的:探讨麝香合剂(SXM)是否与麝香保心丸(SXBXP)一样对急性心肌梗死动物具有保护作用,在急性心肌梗死动物模型上,观察SXM对心肌梗死范围(MIS)的影响,及对缺血区心肌组织中肿瘤坏死因子α(TNF-α)mRNA表达的影响。方法:雄性SD大鼠和雄性Beagle犬,结扎冠状动脉左前降支(LAD)制备心肌梗死模型,24h后,测量大鼠和犬的MIS;检测大鼠缺血区心肌组织中TNF-αmRNA的表达。结果:预先给予SXM可使大鼠和犬的MIS减小,并可明显减少大鼠缺血区心肌组织中TNF-αmRNA的表达。结论:预先给予SXM能够减小急性心肌梗死大鼠和急性心肌梗死犬的MIS,其机制可能与其减少缺血区心肌组织中TNF-αmRNA的表达有关。
OBJECTIVE: To investigate whether SXM has the same protective effect on acute myocardial infarction animals as SXBXP. To observe the effect of SXM on myocardial infarction scope (MIS) in the animal model of acute myocardial infarction. Effect of Tumor Necrosis Factor Alpha (TNF-α) mRNA Expression in Blood Myocardium. METHODS: Male Sprague-Dawley (SD) rats and male Beagle dogs were sacrificed by ligation of the left anterior descending coronary artery (LAD) to establish a myocardial infarction model. After 24 hours, the MIS of rats and dogs was measured. The expression of TNF-α mRNA in the ischemic myocardium was detected. . RESULTS: Pre-administration of SXM reduced the MIS of rats and dogs, and significantly reduced the expression of TNF-α mRNA in the ischemic myocardium of rats. CONCLUSION: Pre-administration of SXM can reduce the MIS in acute myocardial infarction rats and acute myocardial infarction dogs, and its mechanism may be related to the reduction of TNF-α mRNA expression in ischemic myocardium.
