目的探讨杏仁中央核中的N-甲基-D-天冬氨酸(NMDA)型谷氨酸受体对食物和水摄取的影响。方法不锈钢套管植入雄性Sprague-Dawley大鼠一侧杏仁中央核内,NMDA型谷氨酸受体激动剂原型物NMDA,或选择性NMDA受体拮抗剂D()-2-氨基-5-磷酰基戊酸(D-AP-5)微量注射入饱食和含水度正常的大鼠的杏仁中央核内。结果杏仁中央核内注射入8.50,17.00,或34.00 nmol NMDA不改变大鼠摄食量,但是增加注射后0~1 h内不依赖于食物摄取的摄水量(F3,32=3.191,P=0.037)。杏仁中央核内注射入6.34,12.70,或25.40 nmol D-AP-5也不影响大鼠摄食量,但是减少了注射后0~1 h内不依赖于食物摄取的摄水量(F3,28=3.118,P=0.042)。结论杏仁中央核中的NMDA受体可能参与了对摄水而不是摄食的调控。
Objective To investigate the effects of N-methyl-D-aspartate (NMDA) glutamate receptors in central almonds on food and water intake. Methods A stainless steel cannula was implanted into the central almond of male Sprague-Dawley rats. The NMDA-type glutamate receptor agonist (NMDA) or selective NMDA receptor antagonist D (2-amino-5- Phosphoryl pentanoic acid (D-AP-5) was injected in a small amount into the central amygdaloid nucleus of rats with normal satiety and water content. Results The intranuclear injection of 8.50, 17.00, or 34.00 nmol NMDA did not change the rat food intake, but increased the water intake independent of food intake within 0-1 h after injection (F3, 32 = 3.191, P = 0.037) . Intranational CNS injection of 6.34, 12.70, or 25.40 nmol D-AP-5 also did not affect food intake in rats, but decreased food intake independent of 0-1 h after injection (F3,28 = 3.118 , P = 0.042). Conclusion NMDA receptors in central almond nucleus may be involved in the regulation of water intake rather than feeding.
