观察高三尖杉酯碱(HH)对人皮肤瘢痕组织成纤维细胞增殖的影响。利用MTT比色分析法及流式细胞术检测不同浓度HH(μg/ml)作用不同时间对体外培养的人皮肤瘢痕组织成纤维细胞增殖的影响。当HH浓度为0.12μg/ml时即可抑制体外培养的人皮肤瘢痕组织成纤维细胞的生长,0.66μg/ml时即可产生50%的抑制效应,10μg/ml时可完全抑制成纤维细胞的生长,且抑制率随时间的延长而增强,而对正常皮肤组织的成纤维细胞无作用。HH可抑制体外瘢痕组织成纤维细胞的增殖且抑制作用呈剂量时间依赖性。
The effects of homoharringtonine (HH) on the proliferation of human skin scar tissue fibroblasts were observed. The effect of different concentrations of HH (μg/ml) on the proliferation of human skin scar tissue fibroblasts cultured in vitro was detected by MTT colorimetric assay and flow cytometry. When the HH concentration was 0.12 μg/ml, the growth of human skin scar tissue fibroblasts was inhibited in vitro, and 50% inhibition effect was observed at 0.66 μg/ml and completely inhibited at 10 μg/ml. Cell growth, and the inhibition rate increases with time, but has no effect on fibroblasts in normal skin tissue. HH can inhibit the proliferation of scar tissue fibroblasts in vitro and the inhibitory effect is dose-dependent.
