目的研究红花黄色素在大鼠各肠段的吸收动力学特征。方法采用大鼠在体肠段灌流实验,主要从药物浓度、pH值、吸收部位等3方面对红花黄色素的肠段吸收特性进行研究。结果在5~20μg·ml-1浓度范围内药物浓度对吸收速率常数无影响;在pH7.8~5.4内药物吸收不受pH值影响;各肠段的吸收速率常数按空肠、十二指肠、回肠、结肠顺序依次下降,分别为0.1116、0.0753、0.0739、0.0185h-1。结论药物吸收符合一级动力学特征,吸收机制为被动扩散。
Objective To study the absorption kinetics of safflower yellow pigment in rat intestine. Methods Intestinal perfusion experiments were performed in rats. The intestinal absorption characteristics of safflower yellow pigment were studied in terms of drug concentration, pH, and absorption sites. Results The concentration of drug had no effect on the rate constant of absorption in the range of 5-20 μg·ml-1. The absorption of drug was not affected by pH value in pH 7.8-5.4. The absorption rate constants of each bowel segment were jejunum and duodenum. The ileum and colon decreased in turn, and they were 0.1116, 0.0753, 0.0739, and 0.0185h-1, respectively. Conclusion The drug absorption accords with the first-order kinetics and the absorption mechanism is passive diffusion.
