目的:探讨磷酸酯酶2B(PP-2B)的抑制对神经瘤细胞代谢及细胞骨架结构的影响。方法:神经瘤细胞与PP-2B特异性抑制剂共培养,用结晶紫染色和MTT法测定细胞活性,光学显微镜检测细胞形态,免疫细胞染色法检测细胞骨架及PP-2B表达活性。结果:抑制PP-2B可导致细胞代谢明显降低,细胞成活数显著减少;细胞突起消失;微管受损。结论:PP-2B缺陷导致神经瘤细胞代谢和结构双重异常。
Objective: To investigate the effect of inhibition of phosphatase 2B (PP-2B) on the metabolism and cytoskeletal structure of neuroma cells. METHODS: Neuroblastoma cells were co-cultured with PP-2B specific inhibitors. Cell viability was measured by crystal violet staining and MTT assay. The cell morphology was examined by light microscopy, and cytoskeleton and PP-2B expression activity was examined by immunocytochemistry. RESULTS: Inhibition of PP-2B could lead to a significant decrease in cell metabolism, a significant decrease in cell survival, a disappearance of cell protrusion, and damage to microtubules. Conclusion: PP-2B deficiency results in dual abnormalities in metabolism and structure of neuroma cells.
