目的 观察加味真武汤对糖尿病肾病(DN)大鼠IGF-1、肾功能及尿蛋白排泄率的影响,探讨加味真武汤治疗糖尿病的可能作用机制.方法 腹腔注射链脲佐菌素建立大鼠DN模型,随机分为模型对照组、真武汤低剂量组、真武汤高剂量组,另设空白对照组.治疗16周后测定各组大鼠生化指标、尿蛋白排泄率、血清IGF-1、组织中IGF-1定量.结果 模型组大鼠IGF-1、尿蛋白排泄率、血肌酐明显高于空白组,差异有统计学意义(P<0.05);真武汤低、高剂量组与模型组比较各指标明显下降,差异有统计学意义(P<0.05).结论 加味真武汤减少糖尿病肾病大鼠尿蛋白排泄率,作用机制可能与其减少肾脏IGF-1的表达有关.“,”Objective To explore the possible mechanism for the treatment of diabetes by observing the relationship between the IGF-1,renal function and urinary protein excretion rate in rats with diabetic nephropathy after the intervention of flavored Zhenwu decoction. Methods The DN model of rats was established by single dose intraperitoneal injection of streptozotocin and randomly divided into model control group, low dose group,high dose of Zhenwu decoction groups and blank group. The biochemical indexes of rats,urinary protein excretion rate and serum IGF-1 expression were determined after 16 weeks of treatment respectively. Results IGF-1,urinary protein excretion rate and serum creatinine were significantly higher in model control group than those in blank group,the difference was significant (P<0.05). Compared with model control group, each index of low dose group,high dose of Zhenwu decoction groups was significantly lower (P<0.05). Conclusion Flavored Zhenwu decoction reduces diabetic nephropathy proteinuria,and the mechanism may be related to the reduction of renal IGF-1 expression.
