目的研究大蒜素与两性霉素B体外联合应用对白色念珠菌菌丝形成的抑制作用,并检测联合应用时细胞毒性的变化。方法采用微量稀释法测定两类药物联合抗白色念珠菌的作用,采用FICI法对联用结果进行评价。用10%小牛血清RPMI1640培养基诱导菌丝形成,检测大蒜素与两性霉素B联合应用对白色念珠菌芽管率和菌丝形成的影响。通过对细胞形态的观察及MTT法检测联合用药后对人宫颈上皮细胞H8毒性的变化。结果两种药物联用对白色念珠菌表现为协同作用,大蒜素能够加强两性霉素B抑制菌丝形成的能力,联合用药后能够显著降低对H8细胞的毒性。结论大蒜素能够提高两性霉素B抗白色念珠菌的作用,是临床上减少两性霉素B用量,减轻其副作用的最佳选择。
Objective To study the inhibitory effect of combination of allicin and amphotericin B on the mycelial growth of Candida albicans in vitro and the changes of cytotoxicity in combination with allicin. Methods The effect of two kinds of drugs combined with Candida albicans was determined by microdilution method, and the combined results were evaluated by FICI method. The mycelial growth was induced by RPMI1640 medium supplemented with 10% calf serum, and the effect of combination of allicin and amphotericin B on the germ tube formation rate and hyphal formation of Candida albicans was examined. The changes of H8 cytotoxicity in human cervical epithelial cells after combination therapy were detected by observation of cell morphology and MTT assay. Results The combination of the two drugs showed a synergistic effect on Candida albicans. Allicin could enhance the ability of amphotericin B to inhibit the formation of mycelium. The combined treatment could significantly reduce the toxicity to H8 cells. Conclusion Allicin can improve the effect of amphotericin B against Candida albicans and is the best choice to reduce the amount of amphotericin B and reduce the side effects.
