通过盐析、离子交换、凝胶过滤和疏水层析等分离纯化,同步进行抗血管生成和抗肿瘤生长检测,从赤魟(Dasyatis akajei)组织获得强抗血管生成成分福安肽-03(Fuantai-03,FAT-03)的纯品(纯度>95%);使用Edman降解法测定FAT-03 N-末端氨基酸组成和序列,RACE系统技术解析FAT-03的一级结构。结果表明:FAT-03明显抑制鸡胚绒毛尿囊膜血管生成和BALB/c裸小鼠Lewis肺癌的生长及肝转移,且其效果与剂量相关。FAT-03分子的一级结构即其氨基酸组成和序列已被确定。
Separation and purification by salting out, ion exchange, gel filtration, and hydrophobic chromatography, simultaneous anti-angiogenesis and anti-tumor growth assays, and the strong anti-angiogenic component Fuan Peptide -03 obtained from Dasyatis akajei tissue (Fuantai- 03, pure product of FAT-03) (purity > 95%); the N-terminal amino acid composition and sequence of FAT-03 was determined by Edman degradation method, and the primary structure of FAT-03 was analyzed by RACE system technology.
