目的:动态描述药物浓度在体内的不同时间的变化,了解体内药物的行为模式和规律,为个体化治疗和避免药物不良反应提供参考,为开展生理药物动力学模型动态描述奠定基础。方法:使用Simulink动态仿真处理二室模型,建立二室动态模型进行曲线拟合和参数估计,将结果与3P97软件拟合结果进行比较。结果:比较仿真结果与3P97软件拟合结果,两者无显著差异。结论:动态模型可以用于口服给药的二室模型的动态仿真。动态仿真结果更加直观,并纠正了3P97软件的拟合偏差。
OBJECTIVE: To dynamically describe the changes of drug concentration in different time in vivo, to understand the behavioral patterns and laws of drug in vivo, to provide reference for individualized treatment and avoidance of adverse drug reactions, and to lay a foundation for the dynamic description of physiological-pharmacokinetic model. Methods: Simulink was used to simulate the two-compartment model and set up a two-compartment dynamic model for curve fitting and parameter estimation. The results were compared with those of 3P97 software. Results: There was no significant difference between the simulation results and the fitting results of 3P97 software. Conclusion: The dynamic model can be used for dynamic simulation of two-compartment model for oral administration. Dynamic simulation results are more intuitive, and correct the 3P97 software fitting bias.
