目的:探讨上皮型钙黏素(E-cadherin)、β-连环素(β-catenin)和组织蛋白酶D(CathepsinD)在食管鳞状细胞癌(ESCC)中的表达及其与临床病理特征的关系.方法:选取新疆医科大学第一附属医院2003-06/2007-07手术切除并经病理证实的ESCC标本60例,同时选取15例正常食管组织作对照.采用免疫组织化学En Vision二步法检测各组织中E-cadherin、β-catenin及Cathepsin D的表达,分析其与食管鳞状细胞癌临床病理特征的关系.结果:ESCC组织中,E-cadherin和β-catenin异常表达率分别为61.67%和66.67%,CathepsinD的阳性表达率为60%,分别高于正常食管组织(0%,0%,0%,均P<0.01).中低分化组、侵及肌层组及有淋巴结转移组中,E-cadherin和β-catenin的异常表达和CathepsinD的阳性表达均高于高分化组、未侵及肌层组及无淋巴结转移组,差异有统计学意义(P<0.01).ESCC组织中E-cadherin和β-catenin的异常表达呈正相关(r=0.679,P<0.01),CathepsinD的阳性表达与E-cadherin、β-catenin的异常表达呈正相关(r=0.616,r=0.505,P<0.01).结论:ESCC中E-cadherin、β-catenin的异常表达和CathepsinD的阳性表达与肿瘤的浸润和发展有关,联合检测E-cadherin、β-catenin和Cathepsin D有可能成为判断ESCC恶性程度和预后的重要指标.
Objective: To investigate the expression of E-cadherin, β-catenin and Cathepsin D in esophageal squamous cell carcinoma (ESCC) and their relationship with clinicopathological features .Methods: Sixty ESCC specimens surgically removed and pathologically confirmed at the First Affiliated Hospital of Xinjiang Medical University from June 2003 to July 2007 were selected, and 15 normal esophageal tissues were selected as control.Using Envision immunohistochemistry The expressions of E-cadherin, β-catenin and Cathepsin D in each group were analyzed by SPSS 11.5 software.Results: The abnormal expression rates of E-cadherin and β-catenin in ESCC tissues were 61.67% And 66.67%, CathepsinD positive rate was 60%, respectively, higher than normal esophageal tissue (0%, 0%, 0%, all P <0.01) .In the poorly differentiated group, invasive muscle group and lymph node metastasis group , The expression of E-cadherin and β-catenin and the expression of CathepsinD were significantly higher than those in well-differentiated, non-invasive muscular and non-lymph node metastasis groups (P <0.01) The abnormal expression of E-cadherin and β-catenin was positively correlated (r = 0.679, P <0.01). The positive expression of Cathepsin D There was a positive correlation between the expression of E-cadherin andβ-catenin (r = 0.616, r = 0.505, P <0.01) .Conclusion: The abnormal expression of E-cadherin andβ- Invasion and development of the joint detection of E-cadherin, β-catenin and Cathepsin D may be an important indicator to determine the degree of malignancy and prognosis of ESCC.
