该研究建立UPLC-MS/MS测定雪上一枝蒿甲素微透析探针回收率的分析方法,采用浓度差法(增量法,减量法)测定其体外回收率,并考察灌流液的p H,流速,浓度,温度对回收率的影响,以探讨微透析技术用于雪上一枝蒿甲素体内药动学研究的可行性。该试验建立的UPLC-MS/MS分析方法在要求范围内线性良好,色谱的专属性、回收率、精密度等均符合微透析样品的研究要求。增量法和减量法测得回收率无显著差异;相同条件下,探针的体外回收率随着流速的增加而降低,随着温度的升高而明显增加,与探针周围雪上一枝蒿甲素的浓度无关。结果表明,微透析技术可用于雪上一枝蒿甲素药动学研究,反透析法即减量法可用于雪上一枝蒿甲素体内微透析回收率的测定。
In this study, UPLC-MS / MS was used to determine the recoveries of one microtiter microprobe of Artemisia annua on snow. The in vitro recoveries were determined by concentration difference method (incremental method and decrement method) , Flow rate, concentration, temperature on the recovery rate, in order to investigate the feasibility of microdialysis technology for on-snow pharmacokinetics of Artemisia annua. The UPLC-MS / MS analysis method established in this experiment has good linearity within the required range, and the chromatographic specificity, recovery and precision meet the research requirements of microdialysis samples. Under the same conditions, the recovery rate of probe decreases with the increase of flow rate, and increases with the increase of temperature, which is in good agreement with that of Artemisia selengensis A concentration has nothing to do. The results showed that microdialysis can be used to study the pharmacokinetics of pendimethalin in snow. The reverse dialysis method, which is a reduction method, can be used to determine the microdialysis recovery rate of artemisinin in vivo.
