目的探讨Yes相关蛋白(YAP)和细胞周期蛋白D1(CyclinD1)在宫颈上皮内瘤变(CIN)及宫颈鳞状细胞癌(CSCC)中的表达及与CSCC临床病理因素间的关系。方法应用免疫组织化学SP法及RTPCR分别检测105例CSCC、43例CIN和22例正常宫颈组织中YAP和CyclinD1蛋白和mRNA的表达水平。结果 YAP在正常宫颈、CIN和CSCC组织中的阳性表达率分别为22.72%、65.12%和89.52%,三组比较,差异有统计学意义(P<0.05);CyclinD1在正常宫颈、CIN和CSCC组织中的阳性表达率分别为0、11.63%和65.71%,三组比较,差异有统计学意义(P<0.05);YAP和CyclinD1在CSCC中的表达呈正相关(r=0.215,P<0.05)。结论 YAP和CyclinD1的高表达可能与CSCC的发生发展有关,二者有望成为诊断、治疗CSCC的新靶点。
Objective To investigate the expression of Yes-associated protein (YAP) and CyclinD1 in cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma (CSCC) and their relationship with clinicopathological parameters of CSCC. Methods The expressions of YAP and CyclinD1 protein and mRNA in 105 cases of CSCC, 43 cases of CIN and 22 cases of normal cervical tissue were detected by immunohistochemical SP method and RTPCR respectively. Results The positive expression rate of YAP in normal cervix, CIN and CSCC tissues was 22.72%, 65.12% and 89.52% respectively. There was significant difference between the three groups (P <0.05). The positive rate of CyclinD1 in normal cervix, CIN and CSCC (P <0.05). The positive expressions of YAP and CyclinD1 in CSCC were positively correlated (r = 0.215, P <0.05). The positive rates of YAP and CyclinD1 in CSCC were 0, 11.63% and 65.71% respectively. Conclusion The high expression of YAP and CyclinD1 may be related to the occurrence and development of CSCC. Both of them are expected to become a new target of diagnosis and treatment of CSCC.
