目的测定不同采收期木通马兜铃(Aristolochia manshuriensis Kom.)不同部位中马兜铃酸Ⅰ的含量,考察马兜铃酸I的动态累积规律。方法采用HPLC法,色谱柱为DiamonsilTMC18柱(250 mm×4.6 mm,5μm),流动相为乙腈-体积分数0.05%磷酸水溶液,梯度洗脱,检测波长260 nm,进样量10μL,流速1.0 m L·min-1,柱温30℃。结果从5月中旬到6月中旬,木通马兜铃根和叶中马兜铃酸Ⅰ含量均呈上升趋势,6月中旬叶中马兜铃酸Ⅰ含量达到顶峰后呈现下降趋势,而根中马兜铃酸Ⅰ含量继续呈现上升趋势,至7月下旬根中马兜铃酸Ⅰ含量达到峰值,从7月下旬到9月下旬根中马兜铃酸Ⅰ含量呈现下降趋势,而叶中马兜铃酸Ⅰ含量趋于平稳;从5月中旬到9月下旬,木通马兜铃茎、果实中马兜铃酸Ⅰ含量变化处于平稳趋势。结论木通马兜铃中马兜铃酸Ⅰ的分布具有组织特异性和时效性,为马兜铃酸类化合物生合成途径的研究提供重要的科学依据。
Objective To determine the content of aristolochic acid Ⅰ in different parts of Aristolochia manshuriensis Kom. And to investigate the dynamic accumulation of aristolochic acid I. Methods The HPLC method was performed on a Diamonsil TMC18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile-0.05% phosphoric acid by gradient elution. The detection wavelength was 260 nm, the injection volume was 10 μL and the flow rate was 1.0 mL Min-1, column temperature 30 ℃. Results From mid-May to mid-June, the content of aristolochic acid Ⅰ in Aristolochia macrostachya roots and leaves showed an upward trend. The content of aristolochic acid Ⅰ showed a downward trend after mid-June in mid-June, The content of aristolochic acid Ⅰ continued to show an upward trend. The content of aristolochic acid Ⅰ reached the peak value in the end of July, the content of aristolochic acid Ⅰ showed a downward trend from late July to late September, The contents of aristolochic acid Ⅰ tended to be stable. From mid-May to late September, the content of aristolochic acid Ⅰ in the stems and fruits of the Aristolochia atropurpurea was in a steady trend. Conclusion The distribution of aristolochic acid Ⅰ in Corydalis aristolochiae is tissue-specific and time-dependent, which provides an important scientific basis for the study on the pathogengenesis of aristolochic acid.
