目的 :制备包含肉豆蔻酸异丙酯 (IPM )的紫杉醇长效缓释微球 ,并对制备工艺及体内外缓释效果进行评价。方法 :使用改良的单乳溶剂蒸发法制备紫杉醇长效缓释微球。通过分析粒径分布、突释、体外释放及体内的抑瘤效果等指标 ,对其进行评价。结果 :所得微球平均粒径小于 10 μm ,外观圆整 ,具有良好的体外释放曲线。含有30 %IPM的微球体内抑瘤效果明显。结论 :使用改良的单乳溶剂蒸发法制备的微球 ,可以达到缓释效果 ,体外释放4周的微球对小鼠体内的固体肿瘤有良好的抑制效果。
OBJECTIVE: To prepare paclitaxel sustained-release microspheres containing isopropyl myristate (IPM) and to evaluate its preparation process and in vitro and in vivo sustained-release properties. Methods: Paclitaxel sustained-release microspheres were prepared using a modified single-emulsion solvent evaporation method. Through the analysis of particle size distribution, burst release, in vitro release and in vivo anti-tumor effects and other indicators, evaluate them. Results: The average particle size of the obtained microspheres was less than 10 μm. The appearance of the microspheres was round and had a good in vitro release curve. The microspheres containing 30% IPM had obvious anti-tumor effect. Conclusion: The microspheres prepared by the improved single emulsion solvent evaporation method can achieve sustained release effect. The microspheres released in vitro for 4 weeks have good inhibitory effect on solid tumors in mice.
