目的比较低氧诱导因子-1α(HIF-1α)、肿瘤相关巨噬细胞(TAMs)标记抗原CD68和轴突导向因子4D(Sema4D)在胃癌组织和癌旁组织中的表达,探讨三者之间的相关性及临床意义。方法采用免疫组织化学SP法检测86例胃癌组织和癌旁正常组织中HIF-1α,CD68和Sema4D的表达,并分析其与临床病理特征的关系。结果胃癌组织中HIF-1α,CD68及Sema4D蛋白阳性表达率显著高于正常组织(分别为58.1%%VS 23.3%,72.1%VS 23.3%,74.4%VS 29.1%;P均<0.01)。HIF-1α,CD68及Sema4D蛋白与胃癌组织分化类型、TNM分期和淋巴结转移明显有关(P<0.05),且三者表达均呈正相关(r HIF-1α/CD68=0.418,r HIF-1α/Sema4D=0.367,r CD68/Sema4D=0.467,P均<0.01)。结论胃癌组织中HIF-1α,CD68及Sema4D表达明显增高,且三者的表达均呈正相关,三者的联合检测可作为胃癌早期诊断的辅助指标。
Objective To compare the expressions of hypoxia inducible factor-1α (HIF-1α), tumor associated macrophages (TAMs) marker antigen CD68 and axon guidance factor 4D (Sema4D) in gastric cancer tissue and paracancerous tissues, The relevance and clinical significance. Methods The expressions of HIF-1α, CD68 and Sema4D in 86 cases of gastric cancer tissues and adjacent normal tissues were detected by immunohistochemical SP method. The relationship between HIF-1α, CD68 and Sema4D was analyzed. Results The positive rates of HIF-1α, CD68 and Sema4D in gastric cancer tissues were significantly higher than those in normal tissues (58.1% vs 23.3%, 72.1% vs 23.3%, 74.4% vs 29.1%; P all <0.01). The expressions of HIF-1α, CD68 and Sema4D protein were significantly correlated with the differentiation type, TNM stage and lymph node metastasis (P <0.05), and the expressions of HIF-1α / CD68 = 0.418, r HIF-1α / Sema4D = 0.367, rCD68 / Sema4D = 0.467, P <0.01). Conclusion The expressions of HIF-1α, CD68 and Sema4D in gastric cancer tissues were significantly higher than those in normal gastric tissues, and the expression of HIF-1α, CD68 and Sema4D in gastric cancer tissues were positively correlated. The combined detection of the three could be used as auxiliary indicators for early diagnosis of gastric cancer.
