目的了解HIV/AIDS患者外周血白细胞Toll样受体(TLRs)的表达及其在HIV/AIDS疾病过程中的意义。方法应用流式细胞仪检测20例HIV/AIDS患者和15例健康人外周血多型核粒细胞、淋巴细胞和单核细胞表面TLR-1、TLR-2、TLR-4和TLR-6的表达水平。结果 TLR-1、TLR-2、TLR-4和TLR-6在外周血白细胞表面均有表达,与健康对照组比较,HIV/AIDS患者多型核粒细胞TLR-1[(967.44±215.01)%vs(1 159.51±210.17)%,P=0.014]、TLR-2[(1 614.91±346.66)%vs(2 101.87±236.28)%,P<0.001]、TLR-4[(1 735.31±443.28)%vs(2 539.08±842.96)%,P<0.001]和TLR-6[(1 102.96±259.32)%vs(1 357.36±155.49)%,P<0.001]的表达水平显著降低;淋巴细胞TLR-1[(922.09±271.04)%vs(728.19±184.31)%,P=0.024]、TLR-2[(961.81±419.21)%vs(617.72±202.02)%,P=0.008]、TLR-6[(646.93±264.37)%vs(458.50±168.64)%,P=0.037]的表达水平显著升高;而淋巴细胞TLR-4的表达水平两组间差异无统计学意义[(2 067.2±526.32)%vs(2 072.93±259.32)%,P>0.05]。与健康对照组比较,HIV/AIDS患者单核细胞TLR-1[(1 230.67±380.82)%vs(998.86±229.30)%,P=0.014]、TLR-2[(1 091.61±378.23)%vs(549.69±192.99)%,P<0.001]、TLR-4[(2 343.45±724.35)%vs(1 776.67±519.72)%,P<0.001]和TLR-6[(746.58±344.51)%vs(303.19±166.08)%,P=0.004]表达水平均显著升高。结论 HIV/AIDS患者外周血白细胞TLR-1、-2、-4和-6表达水平异常,提示TLRs信号可能在HIV/AIDS发病中起一定作用。
Objective To understand the expression of Toll-like receptors (TLRs) in peripheral blood leucocytes of HIV / AIDS patients and their significance in the process of HIV / AIDS. Methods Flow cytometry was used to detect the expression of TLR-1, TLR-2, TLR-4 and TLR-6 on peripheral blood polymorphonuclear neutrophils, lymphocytes and monocytes in 20 HIV / AIDS patients and 15 healthy controls Level. Results TLR-1, TLR-2, TLR-4 and TLR-6 were all expressed on the surface of peripheral blood leucocytes. Compared with the healthy controls, TLR-1 [(967.44 ± 215.01) TLR-2 [(1 614.91 ± 346.66)% vs (2 101.87 ± 236.28)%, P <0.001], TLR-4 [(1735.31 ± 443.28)%, P = 0.014] The expression of TLR-1 in lymphocytes was significantly lower than that of TLR-6 [(102.296 ± 259.32)% vs (357.36 ± 155.49)%, P <0.001] TLR-2 [(961.81 ± 419.21)% vs (617.72 ± 202.02)%, P = 0.008], TLR-6 [(646.93 ± 264.37)% vs (728.19 ± 184.31)%, P = 0.024] )% vs (458.50 ± 168.64)%, P = 0.037], while there was no significant difference between the two groups in the expression level of TLR-4 in lymphocytes [(2 067.2 ± 526.32)% vs (2 072.93 ± 259.32)%, P> 0.05]. Compared with the healthy control group, TLR-1 [(1 230.67 ± 380.82)% vs (998.86 ± 229.30)%, P = 0.014], TLR-2 [(1091.61 ± 378.23)% vs (746.58 ± 344.51)% vs (303.19 ±), P <0.001], TLR-4 [(2 343.45 ± 724.35)% vs (1776.67 ± 519.72)%, P <0.001] 166.08%), P = 0.004]. Conclusion Abnormal expressions of TLR-1, -2, -4 and -6 in peripheral blood leucocytes of HIV / AIDS patients suggest that TLRs signaling may play a role in the pathogenesis of HIV / AIDS.
