目的:探讨“补肾宁”对氢化可的松法复制肾阳虚模型大鼠卵巢MMP-9mRNA的影响。方法:成年雌性SD大鼠60只,随机分为6组。分别为空白对照组、模型对照组、克罗米芬组、补肾宁低、中、高剂量组。空白对照组每日肌注等容积生理盐水,其余5组,每日下肢肌注氢化可的松注射液,连续11天,制备肾阳虚模型大鼠。11天后按组别分别用药治疗,治疗期为15天。采用原位杂交技术观察各组卵巢局部MMP-9mRNA表达的变化。结果:模型组卵巢MMP-9mRNA的阳性表达明显低于其它各组,其中与补肾宁中、高剂量组比较差异显著(P<0.01);补肾宁中、高剂量组与空白对照组比较MMP-9mRNA表达强度无显著性差异(P>0.05);补肾宁高剂量组与克罗米芬组、补肾宁低剂量组比较有显著性差异(P<0.05)。结论:肾阳虚模型大鼠卵巢MMP-9mRNA表达减弱,可能与卵泡发育异常及排卵障碍有关。补肾宁能够增强肾阳虚模型大鼠卵巢MMP-9mRNA的表达,起到促卵泡发育和促排卵的作用。
Objective: To investigate the effects of “Bushenning” on the expression of MMP-9 mRNA in the ovary of rats with kidney-yang deficiency model by hydrocortisone. Methods: Sixty adult female SD rats were randomly divided into 6 groups. They were blank control group, model control group, clomiphene group, Bushen Ning low, middle and high dose group. In the control group, the same volume of normal saline was intramuscularly injected daily, and the remaining 5 groups were injected with hydrocortisone injection for the lower extremities for 11 consecutive days to prepare a rat model of kidney-yang deficiency. After 11 days, each group was treated with medication and the treatment period was 15 days. In situ hybridization was used to observe the expression of MMP-9 mRNA in the ovary of each group. RESULTS: The positive expression of MMP-9 mRNA in the ovarian tissue of the model group was significantly lower than that of other groups, and there was a significant difference between the high-dose group and the Bushenning medium-dose group (P<0.01). Compared with the blank control group, the MMP- There was no significant difference in the mRNA expression intensity of 9 (P>0.05); there was significant difference between the high-dose Bushening group and the clomiphene group and the low dose group of Bushenning (P<0.05). Conclusion: The decreased expression of MMP-9 mRNA in the ovary of rats with kidney-Yang deficiency may be related to abnormal follicular development and ovulation disorders. Bushening can enhance the expression of MMP-9 mRNA in the ovary of rats with kidney yang deficiency, and play a role in follicle development and ovulation induction.
