目的观察人神经胶质祖细胞移植于小鼠脑内两侧侧脑室周区后的形态结构发育变化。方法将人神经胶质祖细胞移植入出生1天的小鼠脑内两侧侧脑室周区,用免疫荧光组化技术和共聚焦显微镜观察移植6个月后,小鼠脑内人神经胶质祖细胞的分布、分化和形态结构的特点。结果人神经胶质祖细胞移植6个月后成功地在小鼠脑内存活并广泛分布,主要发育成人神经胶质酸性蛋白(glial fibrillary acidic protein GFAP)阳性的星形胶质细胞,表现人星形胶质细胞的形态结构特点,部分神经胶质祖细胞分化成硫酸软骨素蛋白多糖(chondroitin sulphate proteoglycan,NG2)阳性的细胞。结论人神经胶质祖细胞移植小鼠脑内环境中主要分化为GFAP阳性的星形胶质细胞和部分NG2阳性细胞。
Objective To observe the morphological changes of human glial progenitor cells transplanted into the periventricular zone on both sides of the brain in mice. Methods Human glial progenitor cells were transplanted into the periventricular zone of the lateral cerebral ventricle in the brain of one day old mice. Immunofluorescence staining and confocal microscopy were used to observe the changes of human glia Progenitor cell distribution, differentiation and morphological characteristics of the structure. Results Human glial progenitor cells successfully survived and distributed widely in the brain of mice 6 months after transplantation. They mainly developed glial fibrillary acidic protein (GFAP) -positive astrocytes, Astroglial morphological features of some glial progenitor cells differentiate into chondroitin sulphate proteoglycan (NG2) positive cells. Conclusion The glial cells and some NG2 positive cells that differentiate into GFAP positive cells in the brain of human glial progenitor cell transplantation mice.
