目的:基于NF-κB信号通路观察小青龙汤合玉屏风散对变应性鼻炎(AR)大鼠鼻黏膜TNF-α含量和TNF-αmRNA表达的影响,揭示小青龙汤合玉屏风散治疗AR的作用机制。方法:将清洁级健康雄性Wistar大鼠48只按体重采用随机区组法分为6组,即分成正常组、AR模型组、Forskolin(c AMP激活剂)干预组、H89(PKA抑制剂)干预组、PDTC(NF-κB抑制剂)干预组、小青龙汤合玉屏风散组,每组8只。采用卵蛋白全身致敏与局部攻击方法制作AR大鼠模型,观察小青龙汤合玉屏风散组治疗后鼻黏膜TNF-α含量和TNF-αmRNA表达,结果进行统计学分析。结果:模型组TNF-α含量和TNF-αmRNA表达上调,c AMP激活剂Forskolin干预组、NF-κB抑制剂PDTC干预组、小青龙汤合玉屏风散治疗组均能使TNF-α含量和TNF-αmRNA表达下调,而PKA抑制剂H89能上调TNF-α含量和TNF-αmRNA表达。结论:小青龙汤合玉屏风散治疗变应性鼻炎作用机制可能与抑制NF-κB信号通路和兴奋c AMP-PKA信号通路使TNF-α含量和TNF-αmRNA表达下调有关。
OBJECTIVE: To observe the effect of Xiaolonglong Decoction combined with Yupingfeng Powder on the expression of TNF-α and TNF-αmRNA in the nasal mucosa of rats with allergic rhinitis (AR) based on the NF-κB signaling pathway and to reveal the mechanism of Xiaolonglong Decoction combined with Yupingfeng Powder in treating AR. Methods: Forty-eight healthy male Wistar rats were divided into 6 groups according to body weight: normal group, AR model group, Forskolin intervention group, H89 (PKA inhibitor) intervention group Group, PDTC (NF-κB inhibitor) intervention group, Xiaolonglong Decoction combined with Yupingfengsan group, 8 rats in each group. The rat model of AR was established by systemic sensitization and local attack of ovalbumin. The levels of TNF-α and TNF-α mRNA in nasal mucosa were observed after treatment with Xiaolonglong Decoction and Yupingfeng Decoction. The results were statistically analyzed. Results: The levels of TNF-α and TNF-αmRNA in model group were up-regulated. The expressions of TNF-αmRNA and TNF-αmRNA were up-regulated in Forskolin intervention group, PDTC intervention group and Xiaolonglong decoction combined with Yupingfengsan treatment group Downregulation, while PKA inhibitor H89 upregulated the expression of TNF-α and TNF-αmRNA. CONCLUSION: The mechanism of Xiao Qing Long Decoction combined with Yuping Feng San in the treatment of allergic rhinitis may be related to the downregulation of TNF-α and TNF-α mRNA expression by inhibiting the NF-κB signaling pathway and activating cAMP-PKA signaling pathway.
