目的摸索并建立表皮干细胞Oct4基因的非转基因诱导方法。方法采用RT-PCR方法 ,检测经6-氨基己酸刺激48h后表皮干细胞Oct4基因的转录水平。结果 6-氨基己酸能有效激发表皮干细胞Oct4基因的转录。结论 RT-PCR方法检测表皮干细胞Oct4基因转录水平变化的准确度高、重复性好,实验步骤简单,是初步筛选诱导基因转录表达小分子化合物的实用方法 。
Objective To explore and establish a non-transgenic induction method of Oct4 gene of epidermal stem cells. Methods RT-PCR was used to detect the transcription level of Oct4 gene in epidermal stem cells after stimulation with 6-aminocaproic acid for 48 hours. Results 6-Aminocaproic acid can effectively stimulate the transcription of Oct4 gene in epidermal stem cells. Conclusion The RT-PCR method can detect the transcript level of Oct4 gene in epidermal stem cells with high accuracy, good reproducibility and simple experimental procedure. It is a practical method for screening small molecule compounds that induce gene transcription and expression.
