目的:研究雌激素通过子宫内膜癌细胞膜受体非基因效应对细胞内钙离子的调控。方法:17β-雌二醇(E2)和17β-雌二醇偶联牛血清白蛋白(E2-BSA)作用于子宫内膜癌Ishikawa细胞系,在细胞外液分别为细胞培养基、Tyrode’s液和磷酸缓冲液(PBS)的条件下,激光共聚焦显微镜检测雌激素作用后子宫内膜癌细胞胞浆钙离子的浓度变化。结果:细胞外液为培养基时,10nmol/L E2和E2-BSA作用100s内即引起钙离子浓度的增高,E2表现为单峰,E2-BSA表现为双峰;细胞外液为Tyrode’s液时,E2作用后钙离子浓度无改变,E2-BSA引起钙离子浓度快速增高,100s后回落;细胞外液为PBS时,E2作用后钙离子浓度瞬时下降,E2-BSA作用后钙离子浓度快速下降,50s后趋于稳定。结论:雌激素可调节子宫内膜癌细胞内钙离子浓度,细胞外液可影响其调控作用。
Objective: To study the regulation of estrogen on intracellular calcium by non-gene effect of membrane receptor of endometrial cancer cell membrane. Methods: 17β-estradiol (E2) and 17β-estradiol-conjugated bovine serum albumin (E2-BSA) were applied to Ishikawa cell line of endometrial carcinoma. The extracellular fluid was divided into cell culture medium, Tyrode’s solution and The changes of cytosolic calcium concentration in endometrial carcinoma cells were detected by laser scanning confocal microscopy under phosphate buffered saline (PBS) conditions. Results: When the extracellular fluid was used as medium, the concentration of calcium ion was increased within 10s in 10nmol / L E2 and E2-BSA, E2 showed single peak and E2-BSA showed double peak. When the extracellular fluid was Tyrode’s liquid , E2 did not change the concentration of Ca2 +, E2-BSA caused the concentration of Ca2 + increased quickly and then decreased after 100s. When the extracellular solution was PBS, the concentration of Ca2 + decreased instantaneously and the concentration of Ca2 + decreased rapidly After 50s tends to be stable. Conclusion: Estrogen can regulate the intracellular calcium concentration in endometrial cancer cells and the extracellular fluid can influence its regulation.
