本实验用刀豆球蛋白 A-辣根过氧化物酶胶体金(Con A-HRP-G)法定量分析了戊二醛固定后的人外周血白细胞表面受体的分布和密度,可见所有白细胞以及血小板均显示具有 Con A受体。细胞表面平均每μm 周长金标 Con A 复合物的定量分析表明,细胞表面 Con A 受体标记密度因细胞类型而不同。各类血细胞 Con A 受体的标记密度依次为:分叶核中性粒细胞<单核细胞>NK 细胞>杆状核中性粒细胞>嗜碱粒细胞>嗜酸粒细胞>淋巴细胞。小淋巴细胞的密度明显多于大淋巴细胞。NK 细胞表面的受体有极性分布。以上结果说明:1.Con A 受体与细胞功能活性有关,细胞越活跃,则其表面 Con A 受体越多;2.Con A 受体可能与细胞的识别功能有关,如NK 细胞 Con A 受体呈极性分布的现象;3.Con A 受体数目与细胞的成熟程度有关,细胞越成熟,其表面 Con A 受体越多。
In this study, Con A-HRP-G method was used to quantitatively analyze the distribution and density of human peripheral blood leukocyte surface receptors after glutaraldehyde fixation. All the white blood cells And platelets both show Con A receptors. Quantitative analysis of gold-labeled Con A complexes per μm perimeter on the cell surface showed that the Con A receptor labeling density on the cell surface differed depending on cell type. The labeled densities of Con A receptors of all kinds of blood cells were as follows: leaf nucleus neutrophil NK cells> rod-shaped neutrophils> basophils> eosinophils> lymphocytes. The density of small lymphocytes is significantly greater than that of large lymphocytes. The receptors on the surface of NK cells have a polarity distribution. The above results show that: 1. Con A receptor and cell activity, the more active cells, the surface Con A receptors more; 2. Con A receptors may be related to cell recognition function, such as NK cells Con A 3. The number of Con A receptors is related to the degree of cell maturation. The more mature the cells, the more Con A receptors on the surface.
