目的观察咖啡酸苯乙酯(CAPE)对人大肠癌裸鼠皮下移植瘤生长的影响。方法以大肠癌细胞株HCT116为研究对象,对20只BALB/c裸鼠建立人大肠癌皮下移植瘤模型,随机分为对照组和治疗组各10只。治疗组以CAPE5mg/d,观察两组第7、14、21、28天皮下移植瘤生长情况及裸鼠体重的变化;治疗结束时取肿瘤组织及心、肝、肺、肾、肠等行病理组织学检查,TUNEL法检测肿瘤细胞凋亡。结果CAPE对HCT116裸鼠移植瘤的生长有明显抑制作用,与对照组相比,瘤体重量明显降低、体积减小(P<0.01),坏死面积显著增加,凋亡指数明显升高,但对裸鼠体重无明显影响,心、肝、肺、肾、肠等组织未见明显病理学改变。结论CAPE具有抑制人大肠癌裸鼠皮下移植瘤生长的作用,可能与其诱导细胞凋亡有关。
Objective To observe the effect of caffeic acid phenethyl ester (CAPE) on the growth of human colon carcinoma xenografts in nude mice. Methods Human colorectal cancer cell line HCT116 was used as a research object to establish a model of human colorectal cancer subcutaneous xenograft in 20 BALB / c nude mice. The model was randomly divided into control group and treatment group. The treatment group with CAPE5mg / d, observed on the 7th, 14th, 21st, 28th day subcutaneous xenograft tumor growth and nude mice body weight changes; Histological examination, TUNEL assay of tumor cell apoptosis. Results Compared with the control group, CAPE significantly inhibited the growth of xenografted tumor in HCT116 mice, and the volume of the tumor decreased significantly (P <0.01). The area of necrosis increased significantly and the apoptosis index increased significantly No significant effect on body weight of nude mice, heart, liver, lung, kidney, intestine and other organizations showed no significant pathological changes. Conclusion CAPE can inhibit the growth of human colon cancer xenografts in nude mice, which may be related to its induction of apoptosis.
