目的 :建立适宜的小鼠胚胎体外培养体系 ,探讨CO2 气体纯度和培养器皿对小鼠胚胎体外发育的影响。 方法 :采用微滴法培养 2 细胞鼠胚 ,以 4~ 8 细胞鼠胚和桑椹胚形成率作为判断培养效果的指标。 结果 :普通纯度CO2 气体培养组的 4~ 8 细胞形成率、桑椹胚形成率分别为 12 .5 %、0 .0 % ,高纯度CO2 气体培养组分别为6 7.6 %、4 4 .1% ,两组比较差异有统计学意义 (P均 <0 .0 0 1)。国产玻璃器皿培养组的 4~ 8 细胞形成率、桑椹胚形成率分别为 74 .4 %、5 3.5 % ,进口一次性器皿培养组分别为 85 .7%、6 2 .8% ,两组比较差异无统计学意义 (P >0 .0 5 )。结论 :高纯度CO2 气体更适宜鼠胚的体外发育 ;国产玻璃器皿与进口一次性器皿对鼠胚体外发育的效果相近
Objective: To establish a suitable in vitro culture system of mouse embryos to investigate the effects of CO2 gas purity and culture vessels on in vitro development of mouse embryos. Methods: The 2-cell mouse embryos were cultured by microdrop method and the formation rate of 4 ~ 8 mouse embryos and morula embryos was used as an indicator to evaluate the culture effect. Results: The formation rate of 4 ~ 8 cells and the morula formation rate of normal gas CO2 gas culture group were 12.5% and 0. 0%, respectively, while those of high purity CO2 gas culture group were respectively 7.66% and 44.1% The difference between the two groups was statistically significant (P <0.01). The formation rate of 4 ~ 8 cells and the morula formation rate of domestic glassware culture group were 74.4% and 53.5%, respectively, while those of imported disposable vessel culture group were 85.7% and 62.2% respectively The difference was not statistically significant (P> 0.05). Conclusion: High-purity CO2 gas is more suitable for the in vitro development of mouse embryos; Domestic glassware and imported disposable utensils have similar effects on in vitro development of mouse embryos
