目的:探讨黄芩汤对支气管哮喘大鼠外周血淋巴细胞钙调神经磷酸酶(CaN)活性与淋巴细胞增殖及气道炎症影响.方法:选取SPF级Wistar雄性大鼠30只,随机数字表法将大鼠分成三组,模型组(10只)、中药组(10只)与正常组(10只),模型组和中药组大鼠制备哮喘模型,末次给药后将麻醉,取腹部正中切口,采集静脉抗凝血8ml,流式细胞术检测其细胞周期变化,CaN试剂盒检测其活性,ELISA法检测血清IL-4、IL-2含量,并大鼠取右肺叶使用蒸馏水冲洗,甲醛固定,常规包埋切片后行HE染色.结果:模型组大鼠G0/G1期淋巴细胞含量较正常组显著降低,S及G2/M期较正常组显著升高,中药组大鼠G0/G1期淋巴细胞含量较模型组显著升高,S及G2/M期较模型组显著降低,差异均有统计学意义(P<0.05);模型组大鼠CaN活性、IL-4含量较正常组显著升高,IL-2含量较正常组显著降低,中药组大鼠CaN活性、IL-4含量较模型组显著降低,IL-2含量较模型组显著升高,差异均有统计学意义(P<0.05).结论:黄芩汤可使支气管哮喘大鼠淋巴细胞CaN活性及血清IL-4含量降低,抑制淋巴细胞增殖,使气道炎症反应减轻.“,”Objective:To analyze the effect of Huangqi Decoction on the activity of calcineurin (CaN) in peripheral blood lymphocytes and the proliferation of lymphocytes and airway inflammation for asthmatic rats.Methods:Thirty male Wistar rats were selected and randomly divided into three groups:model group (10 rats), Chinese medicine group (10 rats) and normal group (10 rats),.A rat model of asthma was prepared in model group and Chinese medicine group, and anesthetized after the last administration.A midline abdominal incision was made and 8 ml of anticoagulated blood was collected.Flow cytometry was used to detect cell cycle changes.CaN kit was used to test its activity, Serum IL-4, IL-2 were detected by ELISA, content, and the right lung lobe of rats was washed with distilled water and fixed with formaldehyde.HE staining was performed after routine embedding.Results:Compared with the normal group, the lymphocyte content in the G0/G1 phase of the model group rats was significantly lower, and the S and G2/M phases were significantly higher than the normal group.The G0/G1 phase lymphocyte content of the Chinese herb group was significantly higher than that of the model group.The S and G2/M phases were significantly lower than the model group, and the differences were statistically significant (P < 0.05).The CaN activity and IL-4 levels in the model group were significantly higher than those in the normal group, and the levels of IL-2 were significantly greater than those in the normal group.The decrease of CaN activity and IL-4 content in the TCM group was significantly lower than that of the model group, and the IL-2 content was significantly higher than that of the model group, and the difference was statistically significant (P < 0.05).Conclusion:Huangqi Decoction can decrease the activity of CaN in lymphocytes and serum IL-4 in asthmatic rats, inhibit the proliferation of lymphocytes and reduce the airway inflammation.
