目的探讨采用作者前期通过蛋白质组学研究发现的四种鼻咽癌(NPC)患者血清自身抗体来建立多因素判别公式对NPC的临床诊断价值。方法 采用本实验室建立的酶联免疫吸附试验方法(ELISA)分别检测了36例NPC患者、20例其它肿瘤患者及20例健康体检者血清抗细胞角蛋白19(CK19),抗Rho GDP解离抑制因子2(Rho-GDI-2),抗热休克蛋白70(HSP70)和抗亮氨酸氨基肽酶(LAP3)抗体浓度,并对4种抗体浓度采用多因素判别分析方法建立判别函数。结果鼻咽癌患者血清中4种抗体的含量明显高于正常对照组。以4种抗体组成的判别函数对NPC判别的灵敏度为41.7%,特异性为95%,判别正确率为61.8%。结论四种自身抗体组成的多因素判别公式有助于NPC的筛查和诊断。
Objective To explore the clinical diagnostic value of establishing a multivariate discriminant formula for the NPC by using the serum autoantibodies of four NPC patients discovered by the authors in the early stage of proteomics. Methods Serum anti-cytokeratin 19 (CK19), anti-Rho GDP dissociation (Rho) in 36 patients with NPC, 20 patients with other tumors and 20 healthy controls were detected by enzyme linked immunosorbent assay (ELISA) The concentrations of Rho-GDI-2, HSP70 and LAP3 antibody were determined by the method of multivariate discriminant analysis. Results The serum levels of four antibodies in patients with nasopharyngeal carcinoma were significantly higher than those in normal controls. The discriminant function of four kinds of antibodies was 41.7% for NPC, 95% for specificity and 61.8% for discrimination. Conclusion The multi-factor discriminant formula of four autoantibodies contribute to the screening and diagnosis of NPC.
