目的 Notch信号在肠上皮细胞种系分化和引发肠腺瘤和肠癌中起着关键的作用。本文试图探讨Notch信号的配体JAG2在大肠癌中的作用。方法 Real-Time PCR和Western blot分别检测JAG2特异性si RNA对JAG2 m RNA及蛋白表达的抑制率,细胞增殖实验和细胞侵袭实验观察si RNA对细胞增殖及侵袭能力的影响,人类肿瘤转移RT2 Profiler?PCR Array用于筛选JAG2下游靶基因,并应用特异性抑制化合物抑制靶蛋白活性。结果特异性si RNA对JAG2 m RNA及蛋白的表达具有抑制作用(P<0.05),对细胞增殖无明显影响(P>0.05),对细胞侵袭能力明显抑制(P<0.05),而组织蛋白K(CTSK)活性降低可能参与了这一过程。结论 JAG2基因可能对大肠癌细胞的侵袭转移能力起促进作用。
The purpose Notch signaling plays a key role in the germline differentiation of intestinal epithelial cells and in the development of enteric adenomas and colorectal carcinomas. This article attempts to explore the role of Notch signaling ligand JAG2 in colorectal cancer. Methods Real-time PCR and Western blot were used to detect the JAG2 specific si RNA on JAG2 m RNA and protein expression inhibition rate, cell proliferation experiments and cell invasion assay si RNA on cell proliferation and invasion ability, human tumor metastasis RT2 Profiler PCR Array is used to screen JAG2 downstream target genes, and specific inhibitory compounds are used to inhibit target protein activity. Results The specific si RNA could inhibit the expression of JAG2 mRNA and protein (P <0.05), but had no effect on cell proliferation (P> 0.05), but significantly inhibited the invasiveness of cells (P <0.05), while the expression of tissue protein K (CTSK) activity may be involved in this process. Conclusion JAG2 gene may promote invasion and metastasis of colorectal cancer cells.
