目的:研究丹参酮IIA(TanIIA)对佐剂性关节炎(AA)大鼠离体血管的内皮依赖性舒张作用及外周血管假性血友病因子(vWF)表达的影响。方法:建立AA大鼠模型并随机分组,各组连续给药3周后处死大鼠。采用离体血管环灌流讲述观察各组给予去甲肾上腺素(NA)预收缩后加入乙酰胆碱(Ach)的内皮依赖性舒张作用,同时酶联免疫吸附法(ELISA)检测外周血vWF含量。结果:与空白对照组相比,模型组、MTX组血管环应用Ach后对去甲肾上腺素(NA)引起的收缩反应的舒张率显著降低,TanIIA组与空白组相比差异无显著性;模型组vWF水平最高,与空白对照组、MTX组、TanIIA组差异具有显著性,TanIIA组水平最低。结论:AA大鼠主动脉内皮依赖性舒张作用受损机制可能与内皮功能受损有关,TanIIA可以修复内皮细胞,改善RA心血管损伤。
Objective: To investigate the effect of TanIIA on endothelium-dependent vasodilatation and vWF expression in isolated vessels of adjuvant arthritis (AA) rats. Methods: AA rat models were established and randomly divided into groups. Rats in each group were sacrificed 3 weeks after continuous administration. In vitro vascular perfusion was used to observe the endothelium-dependent relaxation of acetylcholine (Ach) precontracted with noradrenalin (NA) and the content of vWF in peripheral blood was detected by enzyme linked immunosorbent assay (ELISA). Results: Compared with the blank control group, the relaxation rate of norepinephrine (NA) -induced contractile response in the model group and the MTX group was significantly decreased after Ach administration, while there was no significant difference between the TanIIA group and the blank group. The model Group vWF highest level, and the blank control group, MTX group, TanIIA group was significant difference, TanIIA group lowest level. Conclusion: The mechanism of endothelium-dependent vasodilation in AA rats may be related to endothelial dysfunction. TanIIA can repair endothelial cells and improve cardiovascular injury.
