目的观察强精片对不育大鼠睾丸生精微环境血-睾屏障(BTB)的影响及可能的机制。方法 70只雄性SD大鼠随机分为空白组(BG组)20只和雷公藤多甙片(GTW)灌胃的造模组50只,灌胃3周后两组各取10只进行模型验证。其余40只造模大鼠随机分为模型组(MG组)和强精片低剂量组(QJPL组)、中剂量组(QJPM组)、高剂量组(QJPH组),灌胃4周后处死分离睾丸,检测相关指标。结果与BG组相比,MG组睾丸组织中FasL、ABP表达量明显上调(P<0.01),而紧密连接蛋白Occludin表达降低(P<0.01)。应用药物强精片干预后,FasL、ABP蛋白在睾丸内表达降低,其中高剂量组更为显著(P<0.01):Occludin蛋白较MG组表达上调,差异具统计学意义(P<0.05)。结论通过上调睾丸组织内FasL、ABP蛋白的表达,改变支持细胞功能,同时抑制Occludin蛋白的表达,破坏BTB完整性,增加其通透性,最终影响生精微环境,这可能是GTW致不育的原因之一。而强精片能够降低睾丸内FasL、ABP表达量,提高Occludin蛋白表达,修复BTB功能,可能是中药制剂改善精子功能的机制之一,但在这一过程中中药介导的具体机制尚需进一步研究。
Objective To observe the effect and its possible mechanism of Qiangjing Tablet on the blood-testis barrier (BTB) in testicular spermatogenic environment of infertile rats. Methods Seventy male Sprague-Dawley rats were randomly divided into two groups: 50 in the blank group (BG group) and 50 in the gavage group of Tripterygium wilfordii (GTW). After 3 weeks of gavage, 10 rats in each group were subjected to model validation. . The remaining 40 model rats were randomly divided into model group (MG group) and Qiang Jing Tablet low-dose group (QJPL group), middle-dose group (QJPM group), and high-dose group (QJPH group), and were killed 4 weeks after intragastric administration. Testes were isolated and relevant indicators were tested. Results Compared with BG group, the expression of FasL and ABP in the testis of MG group was significantly up-regulated (P<0.01), while that of tight junction protein Occludin was decreased (P<0.01). After the intervention of Qiangjingjing Tablets, the expression of FasL and ABP protein in the testis decreased, especially in the high-dose group (P<0.01): Occludin protein was up-regulated compared to the MG group (P<0.05). Conclusion Up-regulation of FasL and ABP protein expression in testicular tissue changes the function of supporting cells, inhibits the expression of Occludin protein, destroys the integrity of BTB, increases its permeability, and ultimately affects the spermatogenic microenvironment, which may be infertility caused by GTW. one of the reasons. Strong membrance tablets can reduce the expression of FasL and ABP in the testis, increase the expression of Occludin protein, and repair BTB function, which may be one of the mechanisms of traditional Chinese medicine preparations to improve sperm function. However, the specific mechanism mediated by traditional Chinese medicine in this process still needs further study. the study.
