目的探讨夏天无注射液调控TNF-α参与大鼠缺血再灌注脑损伤的神经保护作用。方法 SD雄性大鼠随机分为模型组、假手术组、夏天无注射液高剂量组(5 ml/kg)、中剂量组(2.5 ml/kg)及低剂量组(1.0 ml/kg)。改良Zea Longa方法建立大鼠脑缺血再灌注模型。采用在动物麻醉后2.5 h及再灌24 h进行评分;HE染色观察梗死区病理变化;ELISA检测大鼠血液中TNF-α浓度。结果神经功能评分发现,不同剂量夏天无均不同程度提高评分;HE染色发现不同剂量夏天无对脑损伤具有明显改善作用;不同剂量夏天无TNF-α表达量明显下降。结论夏天无提高大鼠脑缺血再灌注后神经功能评分,改善脑组织病理学改变,其机制可能与抑制TNF-α的过度表达有关。
Objective To investigate the neuroprotective effect of summer non-injection of TNF-α in cerebral ischemia-reperfusion injury in rats. Methods SD male rats were randomly divided into 4 groups: model group, sham operation group, non-injection high dose group (5 ml / kg), medium dose group (2.5 ml / kg) and low dose group (1.0 ml / kg) Improved Zea Longa Method to Establish Cerebral Ischemia Reperfusion Model in Rats. The animals were anesthetized 2.5 h and reperfusion 24 h scoring; pathological changes of infarction observed by HE staining; ELISA test for TNF-α in the blood of rats. Results Nerve function score showed that there was no difference in the levels of summer and autumn in different doses. Hematoxylin and eosin (HE) staining showed that no significant difference was found in summer with different doses of summer cerebrospinal fluid. The expression of TNF-α in different doses of summer decreased significantly. Conclusion There is no protective effect on the neurological function after cerebral ischemia and reperfusion in summer, which may be related to the inhibition of TNF-α over-expression.
