目的观察丹参酮ⅡA、苦参碱、川芎嗪对肾上腺嗜铬细胞瘤PC12细胞的抑制作用,并比较3种药物的抗肿瘤活性。方法依据文献分别对丹参酮ⅡA、苦参碱、川芎嗪设定给药浓度,配制9种不同的浓度,对PC12细胞处理24 h,采用MTT测定吸光度值,再根据Origin软件计算细胞的IC50值,比较3种药物对PC12细胞的抗肿瘤作用。结果通过丹参酮ⅡA、苦参碱与川芎嗪对PC12细胞抗肿瘤作用比较,发现丹参酮ⅡA组肿瘤细胞生长明显受抑制。各组IC50值分别为23.21 nmol/mL、11.36μmol/mL、20.32μmol/mL。丹参酮ⅡA、苦参碱与川芎嗪比较,肿瘤细胞生长明显受抑制,差异有统计学意义(P<0.05)。结论与苦参碱及川芎嗪对比,丹参酮ⅡA在低浓度时对肿瘤细胞就有抑制作用,具有显著的抗肿瘤活性。
Objective To observe the inhibitory effects of tanshinone Ⅱ A, matrine and ligustrazine on adrenal pheochromocytoma PC12 cells and to compare the antitumor activities of the three drugs. Methods The concentration of tanshinone Ⅱ A, matrine and ligustrazine were respectively set according to the literature, 9 different concentrations were prepared, the PC12 cells were treated for 24 h, the absorbance was measured by MTT, and then the IC50 value of the cells was calculated according to Origin software. The antitumor effects of three drugs on PC12 cells were compared. Results Tanshinone Ⅱ A, matrine and ligustrazine on PC12 cells antitumor effect and found that tanshinone Ⅱ A tumor cell growth was significantly inhibited. The IC50 values of each group were 23.21 nmol / mL, 11.36 μmol / mL and 20.32 μmol / mL, respectively. Tanshinone Ⅱ A, matrine compared with ligustrazine, tumor cell growth was significantly inhibited, the difference was statistically significant (P <0.05). Conclusion Compared with matrine and ligustrazine, tanshinone Ⅱ A inhibits tumor cells at low concentration and has significant antitumor activity.
