为探讨鸟氨酸脱羧酶(ODC)反义RNA逆转人肺鳞癌L78细胞恶性表型的分子机理,采用核酸杂交、免疫细胞化学染色等方法作研究。结果表明,在ODC反义基因稳定整合、ODC反义RNA高效表达的转染细胞株中,cfosmRNA的表达增强,而人肺癌细胞表面相关抗原的表达明显减少。提示ODC反义RNA抑制多胺合成而引起L78细胞恶性表型逆转,与其对cfos及某些肿瘤相关基因表达的调节作用有关。
In order to investigate the molecular mechanism of the antisense RNA of ornithine decarboxylase (ODC) reversed the malignant phenotype of human lung squamous cell carcinoma L78 cells, nucleic acid hybridization and immunocytochemical staining were used for the study. The results showed that the expression of cfos mRNA was enhanced in the transfected cell lines with stable ODC antisense gene integration and high expression of ODC antisense RNA, while the expression of surface antigens in human lung cancer cells was significantly reduced. It is suggested that the inhibition of polyamine synthesis by ODC antisense RNA may lead to the reversal of the malignant phenotype of L78 cells, which is related to the regulation of c-fos and the expression of some tumor-associated genes.
