DELLA蛋白是赤霉素信号传导通路中一类重要的负调节因子。本研究以珍珠黄杨叶片为试材,利用PCR技术和RACE技术克隆得到珍珠黄杨DELLA蛋白基因Bs GAI1。其c DNA全长2 576 bp,包括1 884 bp完整的ORF,能够编码含有627个氨基酸的蛋白。同时,蛋白相对分子质量为67.39 k D,等电点(p I)为4.99,并且总亲水性平均数为-0.162,预测该蛋白为亲水性蛋白。生物信息学研究表明,Bs GAI1编码蛋白具有DELLA蛋白的典型结构域。实时荧光定量表达分析表明,Bs GAI1基因在珍珠黄杨根、叶、茎和花中都有一定的表达,在根中表达量最低,茎中表达量最高。研究说明,Bs GAI1基因可能在珍珠黄杨茎节间缩短导致矮化的过程中扮演重要角色。
DELLA protein is an important negative regulator of gibberellin signaling pathway. In this study, pearl poplar leaves were used as materials to clone and obtain the DELLA protein gene Bs GAI1 by PCR and RACE techniques. The c DNA is 2 576 bp in length, including 1 884 bp of the complete ORF encoding a 627 amino acid protein. At the same time, the relative molecular mass of the protein was 67.39 kD, the isoelectric point (pI) was 4.99, and the average total hydrophilicity was -0.162. The protein was predicted to be hydrophilic. Bioinformatics studies have shown that Bs GAI1 encoded protein has the typical domain of DELLA protein. The results of real-time fluorescence quantitative analysis showed that the Bs GAI1 gene was expressed in roots, leaves, stems and flowers of P. occidentalis, with the lowest expression level in roots and the highest in stems. Studies have shown that Bs GAI1 gene may play an important role in the process of dwarfing shortening of P. striata.
