目的探讨应用量子点(QDs)荧光探针检测结外NK/T细胞淋巴瘤(ENKTCL)中独立生长因子(Gfi1)蛋白的表达及其与临床预后的关系。方法收集67例确诊为ENKTCL患者的临床病理资料,并进行随访。Gfi1蛋白的表达分别用QDs荧光探针法及免疫组化法检测,并比较2种方法检测的阳性率。应用Kaplan-Meier法及log-rank检验对生存资料进行分析,并比较Gfi1蛋白阳性表达组与阴性表达组的5年总生存率。结果 QDs荧光探针法及免疫组化法检测Gfi1蛋白在ENKTCL中阳性表达率分别为85.07%、73.13%,2者相比差异有统计学意义(P<0.05);2种方法Gfi1蛋白定位相同(细胞核),但荧光探针法的荧光强度较大,亮度较高。Gfi1阳性表达组与阴性表达组的5年总生存率分别为12.90%、54.70%,Gfi1阳性表达组的预后差于Gfi1阴性表达组(P<0.05)。Gfi1蛋白表达是影响生存期的独立预后因素。结论 QDs荧光探针法较免疫组化法能有效识别ENKTCL瘤组织中的Gfi1蛋白,Gfi1蛋白在ENKTCL瘤组织中的表达与ENKTCL的预后有关,Gfi1可作为判断ENKTCL预后的重要指标。
Objective To investigate the expression of Gfi1 in ENKTCL and its relationship with clinical prognosis using fluorescent probes of quantum dots (QDs). Methods Clinicopathological data of 67 patients diagnosed as ENKTCL were collected and followed up. Gfi1 protein expression were detected by QDs fluorescent probe and immunohistochemistry, and compared the positive rate of two methods. Survival data were analyzed by Kaplan-Meier method and log-rank test, and the 5-year overall survival rate of Gfi1 protein positive group and negative expression group was compared. Results The positive rate of Gfi1 protein in ENKTCL detected by QDs fluorescence probe and immunohistochemical method was 85.07% and 73.13%, respectively (P <0.05). The Gfi1 protein localization was the same (Nucleus), but fluorescence probe method of fluorescence intensity, higher brightness. The 5-year overall survival rates of Gfi1 positive group and negative group were 12.90% and 54.70%, respectively. The prognosis of Gfi1 positive group was lower than that of Gfi1 negative group (P <0.05). Gfi1 protein expression is an independent prognostic factor affecting survival. Conclusion QDs fluorescent probe method can effectively identify Gfi1 protein in ENKTCL tumor tissue. The expression of Gfi1 protein in ENKTCL tumor tissue is related to the prognosis of ENKTCL. Gfi1 can be used as an important index to judge the prognosis of ENKTCL.
