目的:优选聚酰胺树脂分离纯化“前愈”总黄酮的工艺条件。方法:采用紫外分光光度法,以总黄酮含量为指标,利用静态和动态结合的方法考察上样质量浓度、pH、树脂药材比、洗脱溶剂的选择及用量等工艺参数,评价聚酰胺树脂纯化总黄酮的行为,确定其最佳纯化工艺。结果:最佳纯化工艺参数为上样液质量浓度1.0 g·m L~(-1),pH 3.0,5 BV水洗脱除杂,洗脱剂50%乙醇,洗脱剂用量4 BV;此工艺条件下,总黄酮纯度由8.89%提高到63.67%,转移率25.51%。结论:采用聚酰胺树脂纯化“前愈”中总黄酮的效果良好,为后期复方中总黄酮部位的活性研究及新剂型的开发提供基础。
Objective: To optimize the polyamide resin separation and purification “Qian Yu ” total flavonoids process conditions. Methods: Ultraviolet spectrophotometry and total flavonoids content were used as indices to evaluate the quality of polyamide resin by static and dynamic methods, such as the concentration of sample, the pH value, the ratio of resin to drug, the choice and dosage of eluting solvent, etc. The total flavonoids behavior, to determine the best purification process. Results: The optimal purification process parameters were as follows: 1.0 g · mL -1 of sample solution, 5 mL of water and 50 BV of eluent, and 4 BV of eluent; Under the conditions, the purity of total flavonoids increased from 8.89% to 63.67%, the transfer rate was 25.51%. Conclusion: Polyamide resin can be used to purify the total flavonoids in Qianqian ’s well, which will provide the foundation for the research on the activity of total flavonoids in later compound and the development of new dosage forms.
