为比较产不同色素古尼拟青霉菌株菌丝体功效成分的差异,本研究对产不同色素古尼拟青霉菌株及对应古尼虫草样品进行试验分析。观察其菌落形态和菌落颜色;用紫外分光光度法测定多糖和D-甘露醇含量;用HPLC法测定虫草素、腺苷、肌苷、胸苷、鸟苷、尿苷6种核苷类成分含量。结果显示,菌落背面绿色的菌株pgjt03,腺苷含量是古尼虫草样品YHCGGB0145的6倍,且虫草素、胸苷、鸟苷、尿苷含量也均达到最高,与它对应的古尼虫草样品YHCGJT0148虫草素、腺苷、鸟苷、尿苷含量均高于其它古尼虫草;而菌落背面浅灰色的菌株pgjt01,核苷类成分含量均低于其他菌株。说明古尼拟青霉产色素与功效成分之间有一定的相关性,古尼拟青霉所产色素颜色越深,功效成分的含量相对越高。通过该研究指出,菌落颜色可作为筛选古尼拟青霉菌株高产功效成分的标志之一,且菌落背面绿色的菌株pgjt03,性状优良,生长较快,可作为潜在的生产菌株。
In order to compare the differences of the functional components of the mycelium of Paecilomyces gunnii in different pigment production, we studied the production of Paecilomyces gonorrhoeae with different pigments and the corresponding samples of Cordyceps militaris. The colony morphology and colony color were observed. The content of polysaccharides and D-mannitol were determined by UV spectrophotometry. The contents of 6 nucleosides of cordycepin, adenosine, inosine, thymidine, guanosine and uridine . The results showed that the green strain pgjt03 on the back of the colony was 6 times higher than the YHCGGB0145 of the genus Cordyceps, and the contents of cordycepin, thymidine, guanosine and uridine were all the highest. The corresponding genus Cordyceps samples YHCGJT0148 The contents of cordycepin, adenosine, guanosine and uridine were all higher than those of other corynebacteria, while the light gray strains pgjt01 and nucleosides on the back of colony were lower than other strains. Explain that there is a certain correlation between the pigments and the efficacy components of the prepared P. gonorrhoeae, and the darker the pigment color of the P. gonorrhoeae, the higher the content of the effective components. According to the study, colony color can be used as one of the markers for screening high-yielding active ingredients of Paecilomyces cinerea, and the green strain pgjt03 on the back of the colony has good traits and rapid growth, and can be used as a potential production strain.
