目的观察常用活血化瘀中药赤芍、当归、红花、川芎、丹参、黄芪及复方当归补血汤、血府逐瘀汤、芎芍胶囊等对鸡胚绒毛尿囊膜(chorioall antoic membrane,CAM)新生血管生成的影响。方法制备CAM模型,分别应用赤芍、当归、红花、川芎、丹参、黄芪及当归补血汤、血府逐瘀汤、芎芍胶囊的含药血清,观察CAM血管生成的情况,同时,设立生理盐水对照组、空白血清对照组、空白对照组及碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)阳性对照组。结果所选药物及bFGF皆有促进CAM新生血管生成的作用,其中,丹参及当归补血汤、血府逐瘀汤、芎芍胶囊等复方在血管生成及血管计数方面,均明显优于生理盐水对照组、空白血清对照组及空白对照组,与阳性对照药bFGF的作用相当;中药复方的作用优于丹参之外的其他单味中药。结论活血化瘀中药丹参及当归补血汤、血府逐瘀汤、芎芍胶囊具有较好的促进CAM新生血管生成的作用。本实验从一个侧面阐明了活血化瘀中药及方剂治疗缺血性疾病的作用机理,同时,研究也体现了中药复方应用的科学依据。
Objective To observe the effects of commonly used traditional Chinese medicines for promoting blood circulation and removing blood stasis, such as red peony, angelica, safflower, Chuanxiong, salvia miltiorrhiza, and radix scutellariae, and compound dangguibuxue decoction, xuefuzhuyu decoction, and decoction capsule on chorioalliantoic membrane (CAM) The effects of neovascularization. Methods The CAM model was prepared. The serum containing erythrocyte, radix sinensis, safflower, Chuanxiong, Salvia miltiorrhiza, and radix scutellariae, and Danggui Buxue Decoction, Xuefu Zhuyu Decoction and Fuling Capsule were used to observe the angiogenesis of CAM. At the same time, the physiology was established. Saline control group, blank serum control group, blank control group and basic fibroblast growth factor (bFGF) positive control group. Results The selected drugs and bFGF all had the effect of promoting angiogenesis in CAM. Among them, Salvia miltiorrhiza and Danggui Buxue Decoction, Xuefu Zhuyu Decoction, Fuling capsule and so on were all significantly better than normal saline in terms of angiogenesis and blood vessel counts. Group, blank serum control group and blank control group had the same effect as positive control drug bFGF; the effect of Chinese herbal compound compound was better than that of other single herbs other than Salvia miltiorrhiza. Conclusion Salvia miltiorrhiza and Danggui Buxue Decoction, Xuefu Zhuyu Decoction and Fuling Capsules have good effects on angiogenesis in CAM. This experiment clarifies the mechanism of action of traditional Chinese medicine and prescriptions for the treatment of ischemic diseases from one aspect. At the same time, the research also reflects the scientific basis for the application of traditional Chinese medicine compound formulas.
