5-烯丙基-7-二氟甲基白杨素诱导肺癌A549细胞生长抑制和凋亡

目的:研究5-烯丙基-7-二氟甲基白杨素(ADFMChR)诱导人肺癌细胞(A549)细胞生长抑制和凋亡的作用.方法:体外培养人肺癌A549细胞,平皿集落形成法测定细胞锚定依赖性生长能力,软琼脂集落形成法测定细胞非锚定依赖性生长能力,流式细胞术(FCM)分析细胞周期和细胞凋亡率;凝胶电泳观察凋亡细胞的基因DNA梯形条带.结果:平皿集落形成法和软琼脂集落形成法结果显示ADFMChR呈剂量依赖性抑制A549细胞生长,FCM分析发现ADFMChR呈剂量依赖性诱导A549细胞凋亡;ADFMChR(10μg/mL)孵育A549细胞48h后,DNA琼脂糖凝胶电泳呈现典型梯形条带.结论:ADFMChR具有抑制人肺癌A549细胞生长和诱导凋亡作用.
