目的 :探讨丙氨酸氨基转移酶胞浆型同工酶 (sALT)在中国人群中多态性分布情况及与其活性的关系。方法 :利用一种不连续电泳缓冲体系的普通琼脂糖凝胶电泳方法 ,对北京地区 486人血清样本进行了ALT酶谱分析 ,检测采用ALT与谷氨酸脱氢酶偶联—MTT呈色反应。结果 :本法最低检出限为ALT4U /L ,sALT各表型可在 40分钟内完成分离 ,sALT各等位基因频率为 :ALT1.0 ,5 5 .86;ALT2 .0 43 4 2 ;ALT3 ,0 .0 0 10 ;ALT1M ,0 .0 0 2 0 ;ALT6.0 .0 0 41;ALT活性在三种常见表型ALT1、ALT2、ALT2 -1间无显著差异 (P <0 .0 5 ) ,而男性的活性显著高于女性 (P <0 .0 1)。结论 :本方法简便、快速、费用低廉 ,适合大规模样本调查。
Objective: To investigate the polymorphism distribution of alanine aminotransferase (sALT) in Chinese population and its relationship with its activity. Methods: A total of 486 human serum samples from Beijing area were analyzed by ALT method using an agarose gel electrophoresis method with a discontinuous electrophoresis buffer system. ALT was detected by ALT and glutamate dehydrogenase conjugate-MTT color reaction . Results: The minimum detection limit of this method was ALT4U / L, and all the phenotypes of sALT could be separated within 40 minutes. The frequencies of sALT alleles were ALT1.0,55.86, ALT2.043 4 2, ALT3 , ALT1M, ALT6.0. 01 41; ALT activity in the three common phenotype ALT1, ALT2, ALT2 -1 no significant difference (P <0. 05 ), While the male’s activity was significantly higher than that of the female (P <0.01). Conclusion: The method is simple, rapid, inexpensive and suitable for large-scale sample survey.
