目的探讨中性鞘糖脂在脑胶质瘤细胞生长调控中的作用。方法用改良的Hakomori方法,提取正常脑组织和脑胶质瘤组织中的中性鞘糖脂,并经柱层析法进一步纯化其中的单一成分红细胞糖苷脂(globoside)和CDH,运用扫描电镜观察它们对胶质瘤细胞生长的影响。结果正常脑组织高表达的红细胞糖苷脂可以抑制体外培养人脑胶质瘤细胞株细胞突起的形成,而脑胶质瘤相关CDH对脑胶质瘤细胞生长无明显影响。结论中性鞘糖脂可以抑制胶质瘤细胞突起的形成,其作用与其结构相关。
Objective To investigate the role of neutral glycosphingolipids in the regulation of glioma cell growth. Methods A modified method of Hakomori was used to extract the neutral glycosphingolipids in normal brain tissues and glioma tissues. The globoside and CDH were purified by column chromatography and observed by scanning electron microscopy. Their effect on the growth of glioma cells. Results The high expression of erythrocyte glycosides in normal brain tissue could inhibit the formation of cell neurites from human glioma cell lines in vitro. However, glioma-associated CDH had no effect on the growth of glioma cells. Conclusion Neutral glycosphingolipid can inhibit the formation of glioma cell protrusion, and its function is related to its structure.
