目的:研究肝癌缺失基因1(deleted in liver cancer-1,DLC-1)在肝癌组织中的表达情况及与肝细胞肝癌临床病理特征的关系。方法:收集50例肝细胞癌患者术后的癌组织及相应癌旁组织的石蜡包埋病理标本及其临床病理资料,另选取10例正常肝组织标本作为对照。应用免疫组织化学法检测上述组织中DLC-1表达情况。结果:DLC-1在肝细胞癌组织中表达阳性率为44%(22/50),显著低于癌旁组织的72%(P=0.005)和正常肝组织的100%(P=0.001),差异有统计学意义(P<0.05)。DLC-1的表达与癌组织病理分化程度、侵袭倾向高低及肿瘤TNM分期有关(P<0.05),而与患者的性别、年龄、HBsAg、肿瘤大小、AFP浓度、纤维包膜及肝硬化程度均无明显相关(P>0.05)。结论:DLC-1在肝细胞癌组织中低表达,且与肿瘤TNM分期、侵袭倾向高低及病理分化程度有关,是肝癌预后的不良因素。
Objective: To investigate the expression of DLC-1 in hepatocellular carcinoma (HCC) and its relationship with the clinicopathological features of hepatocellular carcinoma (HCC). Methods: Paraffin-embedded paraffin-embedded specimens of 50 cases of hepatocellular carcinoma and corresponding paracancerous tissues were collected and their clinicopathological data were collected. Ten specimens of normal liver tissue were selected as control. Immunohistochemistry was used to detect the expression of DLC-1 in these tissues. Results: The positive expression rate of DLC-1 in hepatocellular carcinoma was 44% (22/50), significantly lower than that in para-cancerous tissue (P = 0.005) and normal liver tissue (100%, P = 0.001) The difference was statistically significant (P <0.05). The expression of DLC-1 was correlated with the histopathological differentiation, invasion tendency and TNM stage (P <0.05), but not with the gender, age, HBsAg, tumor size, AFP concentration, fibrous capsule and cirrhosis No significant correlation (P> 0.05). Conclusion: The low expression of DLC-1 in hepatocellular carcinoma tissue is related to the TNM staging, the invasion tendency and the degree of pathological differentiation, which is a negative factor for the prognosis of HCC.
