目的建立链脲佐菌素(streptozotocin,STZ)诱导巴马小型猪糖尿病模型。方法 10头小型猪随机分3组,设为对照组(2头)、实验1组(2头)和实验2组(6头),分别静脉注射生理盐水、120 mg/kg的STZ和150 mg/kg的STZ。在应用STZ后24h内,每小时检测一次血糖,此后每天一次,1个月后处死动物,取胰腺做病理学检查。结果120 mg/kg的STZ可引起一过性糖尿病,血糖在17 d后逐渐恢复。150 mg/kg的STZ,引发3/6头小型猪严重的糖尿病,血糖维持在16.1~35.0 mmol/L之间,不能自发恢复;2/3头小型猪呈一过性高血糖,类似应用120 mg/kg的STZ组动物的变化;此外,还有1/6头动物未发生明显的高血糖。结论巴马小型猪一次性静注120 mg/kg的STZ可引起一过性糖尿病,150 mg/kg的STZ可引起严重的糖尿病,动物对STZ的反应个体差异很大。
Objective To establish a streptozotocin (STZ) -induced diabetes model of parmesan miniature pigs. Methods 10 miniature pigs were randomly divided into 3 groups: control group (2 heads), experimental group 1 (2 heads) and experimental group 2 (6 heads). The animals were injected with normal saline, 120 mg / kg STZ and 150 mg / kg of STZ. Within 24 hours after application of STZ, blood glucose was measured every hour, and once a day thereafter, animals were sacrificed after 1 month, and the pancreas was taken for pathological examination. Results STZ at 120 mg / kg caused transient diabetes and blood glucose recovered gradually after 17 days. STZ at 150 mg / kg triggers severe diabetes in 3/6 piglets with blood glucose maintained at between 16.1 and 35.0 mmol / L and does not recover spontaneously; 2/3 of the minors are transiently hyperglycemic, similar to 120 mg / kg STZ animals; in addition, there are 1 in 6 animals without significant hyperglycemia. Conclusions A single bolus injection of STZ at 120 mg / kg into Bama miniature pigs can cause transient diabetes. STZ at 150 mg / kg can cause severe diabetes and the animals respond differently to STZ.
