采用荧光快速检测 DNA 解螺旋(FADU)方法检测了环境致癌物苯并(a)芘在体外对人外周血白细胞 DNA 的损伤作用。结果显示,经大鼠肝 S_■混合液活化的苯并(a)芘在0.01~100.0μM 剂量范围内可使白细胞 DNA 链断裂明显增加,细胞内剩余双链 DNA 降低24~61%(P<0.05),且存在明显的剂量效应关系。未经活化的苯并(a)芘则未出现明显的 DNA 损伤效应。结果还提示,苯并(a)芘的 DNA 损伤效应是由其活性代谢产物引起的,该 FADU 检测方法同(?)适用于检测能引起 DNA 链断裂的间接致突变物和致癌物。
Fluorescent rapid detection of DNA helices (FADU) method was used to detect the DNA damage of human peripheral blood leukocytes induced by benzo (a) pyrene, an environmental carcinogen. The results showed that in the range of 0.01-100.0μM, the benzo (a) pyrene activated by rat liver S_ ■ mixture significantly increased the DNA strand breaks of leukocytes and decreased the remaining double-stranded DNA by 24-61% (P < 0.05), and there is a significant dose-response relationship. Uninhibited benzo (a) pyrene did not show obvious DNA damage effect. The results also suggest that the DNA damage effect of benzo (a) pyrene is caused by its active metabolite, which is suitable for the detection of indirect mutagens and carcinogens that cause DNA strand breaks.
