目的:探讨类叶升麻苷(Acteoside,AS)对APP/PS1双转基因小鼠老化、焦虑情绪、学习记忆及抗氧化能力的影响。方法:将9月龄的APP/PS1双转基因小鼠随机分为模型组、多奈哌齐组(3mg/kg)、类叶升麻苷低、中、高剂量组(20、40、80mg/kg),每组10~11只,正常对照组为10只同月龄相同背景C57BL/6J小鼠。连续给药135天后,从老化评分、自主活动、O迷宫、穿梭箱实验检测小鼠老化程度、焦虑情绪及记忆能力;采用WST法检测小鼠血清中超氧化物歧化酶(SOD)抑制率,采用比色法测定丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)水平;Luminex法测定了脑组织中BDNF含量。结果:老化评分结果显示,与模型组相比,类叶升麻苷(20、40、80mg/kg)组均能减缓APP/PS1小鼠衰老进程;与模型组相比,类叶升麻苷(80mg/kg)组小鼠躁狂情绪减弱;O迷宫结果显示,类叶升麻苷(20mg/kg)组能缓解小鼠焦虑情绪;类叶升麻苷可升高SOD抑制率和GSH-Px活性,使MDA含量降低;类叶升麻苷(80mg/kg)组有效改善小鼠垂体内BDNF的匮乏状态。结论:类叶升麻苷可通过延缓衰老、改善焦虑情绪,提高学习记忆能力,提高抗氧化能力等途径起到治防老年痴呆的作用。
AIM: To investigate the effects of Acteoside on aging, anxiety, learning and memory and antioxidant capacity in APP / PS1 double transgenic mice. Methods: Nine-month-old APP / PS1 double transgenic mice were randomly divided into three groups: model group, donepezil group (3mg / kg), low, medium and high dose of acteoside group (20,40,80mg / kg) Each group of 10 to 11, the normal control group of 10 same age with the same background C57BL / 6J mice. After 135 consecutive days of administration, the aging degree, anxiety and memory of mice were detected by aging score, voluntary activity, O-maze and shuttle box test. The inhibition rate of SOD in serum was detected by WST method. The levels of malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) were measured by colorimetric method. The content of BDNF in brain tissue was measured by Luminex method. Results: The aging score showed that compared with the model group, the acteoside (20, 40, and 80 mg / kg) groups attenuated the aging process in APP / PS1 mice. Compared with the model group, (80mg / kg) group mice manic mood weakened; O maze results showed that the class of acteoside (20mg / kg) group can relieve anxiety in mice; the class of aconitine can increase the SOD inhibition rate and GSH- Px activity, so that the content of MDA decreased; Acetone (80mg / kg) group effectively improve the lack of BDNF in mice pituitary status. Conclusion: Acetisine can play a role in prevention of Alzheimer’s disease by delaying aging, improving anxiety, improving learning and memory abilities, and improving antioxidant capacity.
