目的建立超声波热水浸提法提取油松松子壳多糖最佳工艺条件。方法利用超声波热水浸提法,苯酚-浓硫酸法检测多糖含量,选择浸提温度、时间、固液比和浸提次数进行单因素梯度实验,再通过L9(34)正交实验优选出油松松子壳多糖提取最佳条件。结果与结论最佳提取条件为浸提温度为60℃,时间为25 min,固液比为1∶3.5,反复浸提2次。该方法所得回收率(n=5)在97.71%~100.67%之间,RSD为1.56%,稳定性较好,为废弃松子壳资源的开发利用建立了一个简单高效的提取方法。
OBJECTIVE To establish the optimum extraction conditions of polysaccharide from Pinus tabulaeformis by ultrasonic hot water extraction. Methods Ultrasonic hot water extraction method, phenol - concentrated sulfuric acid method for polysaccharide content, selection of extraction temperature, time, solid-liquid ratio and the number of single-factor gradient experiments, and then by L9 (34) Pine nutshell polysaccharide extraction of the best conditions. RESULTS AND CONCLUSION The optimum extraction conditions were extraction temperature 60 ℃, time 25 min, solid-liquid ratio 1: 3.5, repeated extraction twice. The recoveries (n = 5) of this method ranged from 97.71% to 100.67% with a RSD of 1.56%. The stability was good and a simple and efficient extraction method was established for the exploitation and utilization of the waste pine shell resources.
